The Melanotan II peptide, or MT-2, is a ring closed by a lactam bridge between aspartic acid and lysine, and it activates MC1, MC3, MC4 and MC5 alike. That breadth is why labs keep it on the shelf: a 2024 review in ACS Pharmacology & Translational Science names MTII among the core tool compounds for validating melanocortin receptor cell lines, solving cryo-EM structures and anchoring high-throughput screens.
The problem is what's actually in the vial, and guessing from forum "reactions" can't settle it. In 2024, forensic toxicologists in Liège confirmed a vial labelled Melanotan II held genuine MT-II at only 30% purity. Milligrams drift too: reports we read for three other sellers' "10 mg" vials measured 9.1, 11.1 and 12.4 mg. Titan Core's MT-2 is third-party tested to 99%+ HPLC purity, and each vial's manufacturer LOT ties to its lab report; if yours isn't posted yet, ask Contact Support before opening. Sold as a lab reagent; never for use in people or animals.
Research Highlights
- Biosensor validation — The genetically encoded FLAREMC sensor gave about a 2.5-fold fluorescence rise to MTII, with a half-maximal response near 12.5 nM (Molecular Metabolism, 2025).
- Antagonist screening — A 2026 macrocycle program used MT-II as its reference agonist, run at EC80–90 to measure each candidate's blocking power (Journal of Medicinal Chemistry).
- Zebrafish melanophore assay — MT-II was the most potent α-MSH derivative tested, and the authors note its 1–2 hour half-life is more than triple α-MSH's 20–25 minutes (International Journal of Molecular Sciences, 2021).
- Hypothalamic neuron recordings — In mouse brain slices, 100 nM MTII excited 37% of histaminergic neurons, 15 of 41 recorded (Molecular Metabolism, 2020).
- Social-behavior models — After 7 days of MT-II, social behavior recovered in maternal-immune-activation mice; normal mice were unchanged (PLoS One, 2019).
- Diet and memory models — After a high-fat diet lasting about 1% of a zebrafish's life, MT-II reversed the recognition-memory and anxiety changes (Biomedicine & Pharmacotherapy, 2023).
